The Mol Switch Project Outcomes


Deliverable 4.1: Combined Magnetic and Optical Tweezer Set-up

CNRS/ENS will produce both a magnetic tweezer and an optical tweezer set-up for measuring the forces required for DNA strand separation of the above palindromic DNA.

Outcomes:
We have built a set-up that allows the simultaneous manipulation of DNA with a magnetic tweezer and the visualization of a single protein tagged with a fluorophore. That set-up needs still to be tested and calibrated. It will then be used to observe the action of a number of DNA translocases, among which will be EcoR124I studied by the groups at Portsmouth and Delft.
